Jack then says”I don’t believe in destiny,” to which Locke replies, “Yes, you do. You just don’t know it yet.” (“Exodus, Part 1”) That last line say alot to me…..but it’s just my opinion.

Most Important line in the series so far….. I think it was said by Locke in “Through The Looking Glass”- When he says “You’re not supposed to do this,” before walking away. I think that this is a very important(yet one of many) aspects to the show. Ben urges Locke to kill Jack but Rousseau knocks Ben out. Jack stands his ground, telling Locke, “You’re done keeping me on this island.” Locke backs down, but tells Jack, “You’re not supposed to do this,” before walking away.
Locke has made similiar comments throughout the show. Another is- After the dynamite incident, Locke asks Jack “Do you think we crashed on this place by coincidence — especially, this place? We were brought here for a purpose, for a reason, all of us. Each one of us was brought here for a reason… The island brought us here. This is no ordinary place, you’ve seen that, I know you have. But the island chose you, too, Jack. It’s destiny.” Jack retorts in a heated moment, asking if Boone’s death was destiny, and Locke says he was a sacrifice. Jack then says: “I don’t believe in destiny,” to which Locke replies, “Yes, you do. You just don’t know it yet.” (“Exodus, Part 1”)

well i think you have covered must of lockes lines, but i like the one that charlie said about locke “no offence mate (to jack) but if there is one person on this island i’d put my absolute faith in to save us all…its john locke”. so i think from this, we can assume that anything locke says or does is important.
i love the “dont tell me what i can’t do” line. thats a classic locke line. :)
to me the most poignant is “you have work to do”

The most important line in the whole series was said by Charlie during the pilot episode, “Where are we?”. Isn’t that essentially what we’ve been trying to do know for 4 years now!
